She was born Danai Jekesai Gurira in Grinnell, Iowa on February 14, to Zimbabwean parents. She was only 5 when her family returned to Zimbabwe. Her father was a university professor; and her mother a college librarian. She has three older siblings and grew up with. Her stage and screen work has been informed by her powerful connection to her African roots.

Gurira returned to the US for university and went to Dominican Convent High School in Zimbabwe. She read psychology at Macalester College in Minnesota before gaining an MFA in acting from the Tisch School of Arts, New York University.
